摘要
目的研究大鼠牙齿移动中整合素αVβ3表达的不同,为研究靶向封闭整合素αVβ3提供时相支持。方法建立大鼠正畸牙齿移动模型,分为成幼鼠两组。拉上颌一侧第一磨牙向近中移动,另一侧为空白对照。分别于0d(未加力时)、加力后3d、1周、3周、4周、6周、8周后将每组5只大鼠处死,行免疫组化技术和计算机图像分析技术比较分析。结果整合素αVβ3两个亚单位的表达峰值在幼鼠出现于第1周,成鼠出现于第3周。3d、1周时幼鼠整合素αVβ3两个亚单位的表达均显著强于成鼠(P<0.05),在3,4,6周和8周时则明显弱于成鼠(P<0.05)。结论成幼鼠整合素αVβ3表达具有不同的时相性变化规律。
Objective To investigate the time expression rules of αvβ3 integrin during experimental tooth movement in the rats,which could provide a basis for studying the targeted blocking integrin αvβ3 . Methods Animal models of orthodontic tooth movement were established in young and adult rats. The first maxillary molar at one side was moved mesially, and the other side was taken as the control side. The rats were killed after 0 d ,3 d, 1,3,4,6, and 8 weeks. The molar-bearing segments were prepared for immunohistochemieal examination and computer image analysis. Results The expression of αvβ3 integrin peaked at week 1 in young rats, and at week 3 in adults. At day 3 and week 1, the expression of both subunits in the young rats was significantly stronger than that in the adult rats( P 〈 0.05 ), while it was significantly weaker at week 3,4,6 and 8 (P 〈 0.05 ). Conclusion The time-expression rules of αv and β3 integrin subunits are different during experimental tooth movement between the young and the adult rats.
