摘要
目的观察骨髓间充质干细胞(MSCs)在糖尿病小鼠体内的归巢和对小鼠糖尿病的治疗作用。方法体外分离和培养出绿色荧光蛋白(GFP)小鼠的mMSCs,经尾静脉将其移植到同品系糖尿病小鼠模型体内。观察GFP-mMSCs在受体体内的分布,检测移植后小鼠血糖、胰岛素、体质量和尿量的变化。结果接受了mMSCs移植的糖尿病小鼠血糖得到一定控制,血清胰岛素增加,体质量恢复,尿量减少,与对照组相比,差异均有统计学意义(P<0.05)。mMSCs移植后30d,在糖尿病小鼠的心脏、肺脏、肝脏、胰腺及肾脏中检测出GFP-mMSCs的分布,但以胰腺和肾脏为多。结论mMSCs在糖尿病小鼠体内有向胰腺归巢的倾向,并对糖尿病有一定的治疗效果。
Objective To observe the nesting an(t the role of therapy of bone-marrowed murine blood glucose , insulin ,body weight and urinary volume were determined respectively after cell transplantation. Results A month after transplantation, the GFP-mMSCs can be detected in lung, heart, liver, kidney and pancreatic tissue of the diabetic mice. The average level of blood glucose decreased and the level of blood insulin increased. There was significant difference between untreated diabetic mice and MSC-treated diabetic mice in body weight and urinary volume. Conclusion The mMSCs have the tendency to home to pancreas of diabetic mice in vivo. MSCs are therapeutically useful in diabetes.
