摘要
用低浓度锌离子从人尿中沉淀尿激酶(UK),锌离子浓度为3mmol/L时,沉淀回收率为87%~93%,用pH10~10.5的20g/L硫酸铵按1∶4(g∶ml)溶解UK沉淀物,UK溶解得率为97%。溶解液用70%饱和度的硫酸铵盐析,UK盐析收率为91%,粗品纯度为4.08×105IU/g,固体效价为3.6×104IU/g,总收率在77%~82%之间。每吨尿(5.00×106IU计)可得UK粗品(3.8~4.1)×106IU。该粗品可用离子交换层析或亲和层析进一步纯化。
Urokinase can be separated from human urine by Zn2+ precipition. The optimal Zn2+ concentration was found to be 3.0mmol/L by which 1.5g wet precipitation was obtained from a litter of fresh human urine with a yield of 87% ̄92%. The precipitation can be dissolved with 20g/L(NH4)2SO4+ammonia (pH 10.0 ̄10.5). The resulting urokinase solution was salted out by (NH4)2SO4 Solution of 70% saturation. The urokinase yield is 91% with purity of 4.08×105 IU/g protein and the solid activity was 3.6×104 IU/g. (3.8 ̄4.1)×106 IU crude urokinase cound be obtained from 1 ton fresh human urine. The overall yield was 77% ̄81%.
