摘要
用乙醇为提取剂,在乙醇浓度80%、温度70℃、料液比1:20、提取时间90 m in条件下回流提取石参根中的总黄酮。以芦丁为标样,NaNO2-A l(NO3)3-NaOH显色体系进行显色,选择波长510 nm,分光光度法测定石参中总黄酮的含量。实验结果表明:石参根中总黄酮含量为1.10 mg/g,加标回收率为99.3%,RSD为2.0%。方法简便、重现性好,准确。
Total flavonoid content in Uraria Crinta Desv. var. macrostachya wall was extracted with ethanol as extractant, under the conditions of 80% ethanol, temperature 70℃, ratio of material to liquid 1:20(w/v ) and extraction time 90rain. Taking by using Rutin as standard sample, and NaNO2 - Al( NO3 ) 3 -- NaOH as chromogenic system,the concentration of total flavonoid was determined by spectrophotometry at the wavelength of 510nm. Re- suits showed the total flavonoid content in Uraria Crinta Desv. vat. macrostachya wall was above 1.10mg/g,the average recovery rate was 99.3% with a RSD of 2.0%. The method was simple, accurate and reproductive.
