宁波地区2008年麻疹病毒分离株H和N基因特征分析 被引量:3

Genetic characterization of H and N gene from wild-type measles viruses isolated in Ningbo in 2008

摘要 目的:分析宁波地区2008年麻疹病毒H和N基因的序列特征。方法:采用RT-PCR扩增2008年宁波地区麻疹分离株血凝素蛋白(H)和核蛋白(N)C末端456 bp核苷酸并进行序列测定,与GenBank中麻疹病毒各基因型参考株比较并分析毒株变异情况。结果:共分离到11株麻疹病毒,均为H1a基因亚型。与H1a参考株China93-2的H和N基因的氨基酸同源性分别为98.5%-98.7%和96.1%-98.7%;与中国疫苗株S191的H和N基因的氨基酸同源性分别为95.0%-95.5%和86.8%-89.5%。结论:宁波地区2008年流行的麻疹病毒均为H1a基因亚型,与中国疫苗株S191在基因特征上存在明显差异。 Objective: To investigate the genetic characterizations of H and N gene of measles viruses currently prevalent in Ningbo city.Methods:Measles viruses were isolated from throat swab specimens collected from suspected measles cases and the H and 456 bp fragments of C terminus of nucleoprotein(N) gene were amplified by RT-PCR.Sequence analysis was conducted of all 11 virus strains while compared with other measles virus strain published in GenBank.Results:All of the 11 measles isolates belonged to genotype H1a.The homogeneities of amino acids of H and N gene were respectively 98.5%~98.7% and 96.1%~98.7% while compared to the representative H1a genotype strain China93-2,the datas were 95.0%~95.5% and 86.8%~89.5% while compared to Shanghai 191.Conclusion:Genotype H1a measles virus had been proven to have circulated in Ningbo,China in 2008.There are obvious differences on genetic characteristics between the isolates and Shanghai 191.
