RP-HPLC法同时测定十味活血丸中阿魏酸、橙皮苷和5-O-甲基维斯阿米醇苷的含量

RP-HPLC simultaneous determination of ferulic acid,hesperidin and 5-O-methylvisammioside in Shiwei Huoxue pills

目的:建立同时测定十味活血丸中阿魏酸、橙皮苷和5-O-甲基维斯阿米醇苷3种活性成分含量的RP-HPLC法。方法:采用Diamonsil C18(200mm×4.6mm,5μm)色谱柱;以甲醇-0.05%醋酸-四氢呋喃(34∶65∶0.2)为流动相;流速1.0mL.min-1;柱温35℃;检测波长320nm。结果:阿魏酸、橙皮苷和5-O-甲基维斯阿米醇苷浓度分别在1.80～18.0,40.0～400,3.00～30.0μg·mL-1范围内与峰面积呈良好线性关系(r>0.9995,n=6);方法平均回收率(n=9)分别为101.6%,99.9%,98.0%。结论:本法简便、准确,重复性好,可为十味活血丸质量评价提供依据。

Objectives： To develop a RP-HPLC method for simultaneous determination of ferulic acid, hesperidin and 5-O-methylvisammioside in Shiweihuoxue pills. Methods： The analysis was carried on a DiamonsilC18（200 mm × 4.6 mm, 5 μm）column with the mobile phase methanol-water containing 0.05%acetic acid-tetrahydrofuran（34 ： 65 ： 0.2） at a flow rate of 1.0 mL·min-1. The UV detection wavelength was set at 320 nm and the column temperature was 35 ℃. Results： The linear ranges of ferulic acid, hesperidin and 5-O-methylvisammioside were 1.80-18.0, 40.0-400, 3.00-30.0 μg·mL-1 （r〉0.999 5, n = 6）, respectively. The average recoveries of Ferulic acid, Hesperidin and 5-O- Methylvisammioside were 101.6%, 99.9% and 98.0% （RSD〈3.0%, n = 9）, respectively. Conclusions： This method is suitable to control the quality of Shiweihuoxue pills, which provides the advantages of simplicity, accuracy and reproducity.
