摘要
从方法学上探讨了样品浓度、溶剂、pH、盐浓度对白蛋白多肽清除DPPH·能力的影响,以及白蛋白多肽、VC、GSH与DPPH反应随着时间推移的动力学关系,并对其EC50进行比较。结果表明,最佳测定条件为以50%乙醇为溶剂,浓度范围为1~7mg/mL,反应时间50min,运用此法测定的清除率在16%~80%之间,具有较高的灵敏度和准确性。白蛋白多肽:EC50=3·64mg/mL,VC:EC50=3·38×10-3mg/mL,GSH:EC50=0·053mg/mL。
In this article the free-radical scavenging effects of the different concentrations of the ovalbumin peptide, different solvents, different pH and different salt concentrations by DPPH· method was studied.The results showed that the optimum assay range was 1-7mg/mL and optimum time was 50min.The scavenging scope was between 16%-80% .The sensitivity and accuracy of this method were high.The kinetics properties of scavenging free-radical were introduced,The results showed that the kinetics properties of ovalbumin peptide were similar to the GSH,but different from Vc.Comparison of the ECho of the ovalbumin peptide to GSH,Vc showed that the order of EC50:Vc 〉 GSH 〉 the ovalbumin peptide,the ovalbumin peptide' s EC50 = 3.64mg/mL,Vc' s EC50 = 3.38 × 10^-3mg/mL, GSH' s EC50= 0.053 mg/mL, respectly.
