摘要 探讨草石蚕多糖提取及纯化工艺,通过单因素和正交实验摸索提取条件,应用超滤技术进行纯化,最终确立了草石蚕多糖最佳提取工艺:提取温度为60℃,时间为1h,料液比为1∶30,在此条件下草石蚕多糖的提取率为53·86%,草石蚕提取液经截留分子量为3kD的中空纤维膜超滤处理后,其透过率为76·2%,透过液多糖纯度达77·0%,用薄层层析(TLC)鉴定其透过液的主要成分为水苏糖。 In order to study the extraction and purification technology of polysaccharide from Stachys seiboibi Miq, the conditions were explored by the single factors and orthogonal tests, ultrafiltration was applied and finally the optimum extraction technology of polysaccharide from Stachys seiboibi Miq was established. It was the best temperature 60℃,the total time 1h, the solid-liquid ratio 1:30.The extraction yield could be up to 53.86%.Besides, for the membrane with a cut-off molecular mass of 3kD, the transmittance of polysaccharide was 76.2%, and the purity of polysaccharide filtration fluid of 77.0% could be reached.The main component of polysaccharide filtration fluid proved to be stachyose by TLC.
