摘要
目的:制备水飞蓟油脂质体并建立其质量评价方法。方法:采用薄膜分散法,以包封率为检测指标,利用正交试验法优选处方和制备工艺;考察优选处方所制脂质体形态与粒径,并采用透析法结合紫外分光光度法测定其包封率。结果:最佳处方为制备的脂质体为药脂比1:15,卵磷脂和胆固醇质量比为5:1,磷酸盐缓冲溶液的浓度为1/1。脂质体呈球状小囊泡,外观圆整,粒径范围(149.2±59.5)nm,包封率为95.67%。结论:优选处方和制备工艺稳定可行,可为开发其新剂型提供参考。
Objective To prepare silybum marianum gaertn seed oil liposomes and establish a quality evaluation method thereof. Methods The liposomes were prepared by thin-film hydration method. The entrapment ratio as quota with encapsulation efficiency of liposomes,the optimal formation and preparation process were selected by an orthogonal design test. The encapsulation efficiency of liposomes was determined by UV spectrophotometry combined with the use of dialysis methods after liposomes were being dialyzed. Transmission electron microscopy was performed for examining the appearances of liposomes and the diameter of the particles. Results As the best prescription for the preparation of liposomes, the best weight radio of S.M.G seed oil to lecithin is 1:15,1ecithin to cholesterin is 5:1 and the concentration of phosphate buffer solution is 1/1.The encapsulation efficiency and the diameter range of S.M.G seed oil liposomes were 95.67% and (149.2±59.5)nm,respectively.The average recovery was (98.62±1.45)%(RSD=1.47%)Conclusions The selected formulation and preparation of S.M.G seed oil liposomes is practicable.
