黑曲霉MA-56β-甘露聚糖酶的生产条件研究 被引量:2

摘要 本文通过Plackett-Burman设计及单因素试验,研究了影响黑曲霉菌株MA-56生产β-甘露聚糖酶的主要因子,并优化了影响β-甘露聚糖酶固体生产的因素。结果表明,硫酸亚铁、牛肉膏、磷酸二氢钾和玉米浆粉4个因素对β-甘露聚糖酶生产影响显著,可信度大于90%,但均为产酶负影响因子;尿素、氯化钙和硫酸镁为产酶的正影响因子,但影响不显著。单因子试验表明,黑曲霉菌株MA-56产β-甘露聚糖酶的最适培养基条件为:麸皮8g,豆粕2g,魔芋粉0.1g(以10g干基为标准)。当固形物与加水比10∶9,接种孢子悬浮液2.5mL(以一支菌种斜面加30mL无菌水为标准),300mL三角瓶中装量7g培养基,发酵56h时,产酶量达到2.01×105U/g。 Plackett-Burman design and the single factor design were used to evaluate the nutritional requirements of A spergillus niger MA-56 β-mannanase production.The results showed:FeSO4,beef extract, KH2PO4 and corn slurry powder repressed the β-mannanase production significantly, and urea, CaCl2 and MgSO4 induced the β-mannanase production weakly.The optimal culture medium were 8 g wheat bran,2 g bean powder,0.1 g konjak powder (counted by 10 g dry medium ).The optimum cultural conditions for β-mannanase production were as follows:the ratio of solid substrate and water was 10:9,2.5 mL spore suspension was inoculated in medium,7 g medium (calculated by dry matter)was loaded in 300 mL shake flask.Under that condition, β-mannanase production by A spergillus niger MA-56 can reach 2.01-10^5 U/g after 56 h fermentation.
