慢性荨麻疹患者外周血单个核细胞TLR2、TLR4表达及其与IL-5的相关性

Expression of TLR2、TLR4 on peripheral mononuclear cells with the linkage to IL-5 in patients with chronic urticaria

目的:观察慢性荨麻疹患者单个核细胞表面TLR2、TLR4表达及其与血清IL-5的相关性;初步探讨TLR2、TLR4在慢性荨麻疹发病机制中的作用,为临床治疗提供新思路。方法:采用流式细胞仪检测49例慢性荨麻疹患者外周血单个核细胞TLR2及TLR4受体表达情况;用双抗夹心ELISA方法检测患者血清中IL-5浓度,与30例同期正常对照。结果:慢性荨麻疹患者外周血单个核细胞TLR2表达均明显高于正常对照,而TLR4表达与正常对照无明显差异(P>0.01),血清中IL-5浓度明显高于正常对照,TLR2表达与血清IL-5浓度呈正相关,TLR4表达与血清IL-5浓度呈无相关。结论:研究提示TLR2可能在慢性荨麻疹的发病中起重要作用,其机制可能是通过上调TLR2表达促进IL-5分泌来介导变态反应。

Objective ：To discuss the role of Toll- like receptor （TLR2）／TLR4 in chronic urticaria and to provide a new method to cure the chronic urticaria, the expression of TLR2 on peripheral blood rnononuclear cells and its correlation with concentration of IL -5 in the patients with the chronic urticaria were investigated. Methods：The expression of TLR2 ／TLR4 mRNA on peripheral blood mononuclear cells in 49 cases of the chronic urticaria was examined by flow cytometry, the content of IL -5 in serum was examined by ELISA. Blood from thirty cases of healthy people was used as control group. Results：The expression of TLR 2 and the serum level of IL -5 of patients were significantly higher than those in control group （P 〈 0.01 ）, and the expression of TLR2 was positively correlation with level of IL -5. Conclusion：TLR2 may play a critical role in the mechanism of the chronic urticaria, which may upregulate the secretion of inflammatory factors to induce onset of chronic urticaria.
