槲皮素和乙酸龙脑酯对细菌脂多糖诱导流产小鼠子宫组织IFN-γ/IL-4和巨噬细胞的影响 被引量:6

摘要 目的探讨小鼠子宫组织中巨噬细胞在早期胚胎丢失中的意义,研究槲皮素和乙酸龙脑酯对细菌脂多糖(LPS)诱导流产孕小鼠的保胎效果,和对母胎界面免疫平衡的调节作用。方法选用LPS尾静脉注射(0.10μg/鼠),制造小鼠流产模型,孕4~7d分别口服不同保胎药物,用药前后检测各组(n=10)小鼠子宫组织干扰素γ/白细胞介素4(IFN-γ/IL-4)的含量和巨噬细胞数量的变化。结果LPS促流产后,子宫匀浆中IFN-γ含量升高,IL-4含量降低,IFN-γ/IL-4的比值升高,与对照组比较差异极显著;孕小鼠子宫壁巨噬细胞数量显著增多。预先口服槲皮素和乙酸龙脑酯可不同程度地抑制LPS的作用,其中以槲皮素和乙酸龙脑酯联合使用组的保胎效果最为明显,IFN-γ/IL-4的比值基本达到正常水平,巨噬细胞数量降至16.199±0.802,较LPS流产模型组差异极显著。结论小鼠子宫组织中IFN-γ/IL-4的比值升高和巨噬细胞数量增多与早期胚胎丢失关系密切,槲皮素和乙酸龙脑酯能调节子宫局部的免疫微环境,从而起到一定的促孕保胎作用。 Objective To investigate significance of interferon-T/interleukin-4(IFN-γ/IL-4) and macrophages in uterus in early embryo loss (or resorption), and to elucidate the anti-abortive effect and the immunological modulation at maternal-fetal interface with quercetin and bornvl acetate. Methods Lipopolysaccharide (LPS)(0.10μg/mouse)was injected via tail vein in order to induce abortion in 7-day-gestation mice which received quercetin and borud acetate at days 4-7 of gestation. Levels of IFN-γ and IL-4 in uterus lysate superuatant were measured using enzyme-linked immuno-absorbent assay (ELISA), and uterine macrophages of each group ( n = 10) were detected by immunohistochemistry. Results The amount of macrophages was much higher in the uteri of LPS-induced abortion mice than that in control mice. The abortion rate of mouse declined to certain level. The therapy of quercetin combined with boruyl acetate reversed LPS-induced abortion. Conclusion The increase of IFN-γ/IL-4 and the amount of maerophages in the LPS-treated mouse uterus is associated with the embryo loss, and quercetin and boruyl acetate have the anti-abortive effect through modulation of maternal-fetal interface immunity balance.
