百草枯中毒大鼠肺组织血栓调节蛋白和内皮细胞蛋白C受体表达的变化及二巯丙磺钠的作用 被引量:5

Expression of thrombomodulin, endothelial protein C receptor in lung tissue of acute paraquat poi- soned rats and intervention of sodium dimercaptopropane suffonate

摘要 目的观察急性百草枯(paraquat,PQ)中毒大鼠肺组织中血栓调节蛋白(thrombomodulin,TM)及内皮细胞蛋白c受体(endothelialprotein Creceptor,EPCR)mRNA表达的变化及二巯丙磺钠(Na-DMPS)的干预作用。方法80只雄性SD大鼠随机分为4组:正常对照组(8只);Na-DMPS对照组(8只,腹腔注射200mg/kgNa-DMPS);PQ染毒组,32只,腹腔注射体积分数为1%的PQ溶液(20mg/kg)1;Na-DMPS保护组32只,腹腔注射200mg,kgNa—DMPS15min后再腹腔注射体积分数为1%的PQ溶液(20mg/kg)]。检测PQ染毒组及Na—DMPS保护组染毒后6h及1、3、7d肺组织巾TMmRNA、EPCRmRNA的表达,并观察大鼠肺组织的病理学变化。结果PQ染毒组染毒后3d内炎性细胞(如中性粒细胞、淋巴细胞等)浸润明显,伴充血水肿;Na—DMPS保护组炎性细胞浸润较PO染毒组少,充血水肿轻。与对照组相比,PQ染毒组及Na—DMPS保护组大鼠肺组织中TMmRNA、EPCRmRNA的表达增强,在6h达高峰,1d开始下降,7d降至正常水平。Na—DMPS保护组6h及1dTMmRNA表达分别为1.071±0.097、1.055±0.051,与PQ染毒组比较,明显降低,差异有统计学意义(P〈0.05或P〈0.01);Na—DMPS保护组6h及1dEPCRmRNA表达分别为0.678±0.005、0.650±0.007,与PO染毒组比较,明显降低,差异有统计学意义(P〈0.05或P〈0.01)。结论大鼠急性PO中毒后肺组织中TMmRNA、EPCRmRNA的表达升高,预防性应用Na—DMPS后,TMmRNA、EPCRmRNA的表达降低。 Objective VTo observe the expression and effect of thrombomodulin (TM)mRNA and en- dothelial protein C receptor(EPCR) mRNA in lung tissue of acute paraquat poisoned rats, and intervention of sodium dimercaptopropane sulfonate (Na-DMPS). Methods Eighty male SD rats were randomizedly divided into four groups: the normal control group (n=8), the Na-DMPS control group (n=8, administered with 200 mg/kg Na-DMPS intraperitoneally), the PQ group (n=32, administered with 20 mg/kg I%PQ intraperitoneally), the NA-DMPS protected group (n=32, administered with 200 mg/kg Na-DMPS intraperitoneally before with 20 mg/kg 1%PQ). The expression of TM mRNA and EPCR mRNA in the PQ group and the Na-DMPS protected group was evaluated at the six hour, on the first, third and seventh day. Results The expression of TM mRNA and EPCR mRNA in lung tissue of poisoned rats, was significantly increased and reached the peak at the six hour, was decreased slowly on the first day, and returned to normal level on the seventh day. In the Na-DMPS protected group, at the six hour and on the first day, the expression of TM mRNA ( 1.071±0.097, 1.055±0.051 ) was less than that in the PQ group (P〈0.05 or P〈0.01 ). EPCR mRNA (0.678±0.005), (0.650±0.007)at the six hour and on the first day in the Na-DMPS protected group was less than that in the PQ group (P〈0.05 or P〈 0.01 ). Conclusion The expression of TM mRNA and EPCR mRNA of rats after PQ intoxication is increased, and can sianificantlv be decreased after administered with Na-DMPS.
