姜黄素对丙烯酰胺细胞毒性和遗传毒性的保护作用

摘要 目的体外研究姜黄素对丙烯酰胺(AA)所致人肝癌HepG2细胞细胞毒性和遗传毒性的保护作用。方法AA处理组(细胞毒性实验设5,10,20,40和80mmol/L5个剂量,微核试验设1.25和2.50mmol/L2个剂量),姜黄素保护组设0.63、1.25和2.50μg/mL3个剂量组,噻唑蓝法观察姜黄素对AA所致细胞毒性的保护作用;微核试验观察姜黄素对AA所致遗传毒性的保护作用。结果随着AA浓度的升高,HepG2细胞的存活率逐渐下降,与相应浓度AA处理组比较,2.50μg/mL姜黄素保护组的细胞存活率升高,差异有统计学意义(P〈0.05);1.25、2.50mmol/LAA组HepG2细胞微核率分别为(41.3%±50%)和(46.0%±4.0‰),明显高于溶剂对照组,差异有统计学意义(p〈0.05)。2.50μg/ml姜黄素保护组细胞微核率明显低于1.25和2.50μg/mlAA组,差异有统计学意义(P〈0.05)。结论姜黄素对AA所致HepG2细胞的细胞毒性和遗传毒性具有保护作用。
