摘要
目的探讨活性氧(ROS)在TNFα刺激人脐静脉内皮细胞(HUVEC)胞浆游离钙水平变化中的作用。方法用氧化-还原敏感的荧光探针2,7-Dichlorofluorescein(DCF)检测细胞内氧化-还原水平。用激光共聚焦显微镜检测胞浆游离钙浓度。结果TNFα(200U/ml)处理24h后,HUVEC胞内ROS水平和游离钙浓度均明显升高;NAC(20mmol/L)预处理可明显抑制胞浆内游离钙浓度(P<0.01)。结论ROS部分介导了TNFα对胞浆内游离钙水平的诱导作用。
Objective To investigate the roles of reactive oxygen species(ROS) on the eytosolic free calcium(FECa^2+]i) in the endothelial cells stimulated by TNFα. Methods The ROS production in the cells was measured by 2,7 - Dichlorofluorescein(DCF) diacetate as probe; The cytosolic free calcium (F[Ca^2+ -]i) was detected by confocal fluorescence microscope. Results TNFα(200 U/ml) treatment caused an increase in the cytosolic free calcium concentration and the ROS production in HUVECs, which was markedly reduced by pretreatment with NAC(20 mmol/L). Conclusions ROS may play a major role in the TNFα-induced upregulation of F[Ca^2+]i.
