S-腺苷蛋氨酸和叶酸抑制同型半胱氨酸诱导的海马神经细胞凋亡 被引量:5

STUDY ON INHIBITION OF S-ADENOSYL METHIONINE AND FOLATE ON HOMOCYSTEINE-INDUCED APOPTOSIS OF HIPPOCAMPAL NEURONS

摘要 目的研究S-腺苷蛋氨酸和叶酸抑制同型半胱氨酸诱导的海马神经细胞凋亡作用及其相关机制。方法用同型半胱氨酸、S-腺苷蛋氨酸和叶酸处理体外培养的海马神经细胞,观察神经细胞的凋亡情况,以及凋亡相关蛋白的表达变化。结果用250μmol/L同型半胱氨酸作用4h可使神经细胞凋亡率显著升高,并呈时间依赖性。甲基供体S-腺苷蛋氨酸和叶酸可显著抑制同型半胱氨酸引起的神经细胞凋亡。同型半胱氨酸可显著促进凋亡相关蛋白P53、Bax的表达,抑制Bcl-2的表达。S-腺苷蛋氨酸和叶酸可抑制同型半胱氨酸引起的凋亡相关蛋白表达的改变。结论S-腺苷蛋氨酸和叶酸可显著抑制同型半胱氨酸引起的神经细胞凋亡,凋亡相关蛋白表达的变化可能是S-腺苷蛋氨酸和叶酸作用的分子机制之一。 Objective To observe the effects of S-adenosyl methionine (SAM) and folate inhibiting homocysteine (Hcy)-induced apoptosis of hippocampal neurons in vitro and the related molecular mechanisms. Method We examined the consequences of treatment of hippocampal neurons with Hcy, SAM and folate and detected the expression of apoptosis related protein. Results Exposure of primary hippocampal neurons to Hcy (250 μmol/L) resulted in apoptosis, and the apoptosis was time dependent. SAM and folate could significantly repress Hcy induced neuron apoptosis. Hcy significantly increased the expression of P53 and Bax while decreased the expression of Bcl-2. SAM and folate could inhibit the changes of apoptosis related protein expression induced by Hcy. Conclusion SAM and folate repress the Hcy induced neuron apoptosis by inhibiting the expression changes of the apoptosis related protein.
