复方枸杞子口服液对孕鼠致畸性的研究

Study of teratogenicity of compound medlar oral solution in pregnant rats

目的观察复方枸杞子口服液经灌胃对大鼠的生殖毒性和胚胎致畸作用。方法SPF级Wistar大鼠随机分成空白对照组、复方枸杞子口服液高、中、低剂量组、阿司匹林组(阳性对照组)。灌胃量按10.0mL/kgBW计算,空白对照组灌胃给予同体积的注射用水。孕鼠于7～16d每天灌胃给药,妊娠20d处死,分析其胚胎发育指标与胎仔发育指标,检查有无外观畸形和骨肋畸形。结果复方枸杞子口服液对孕鼠生殖能力无影响;对胎鼠体质量、身长、胎盘重量均无影响;胸骨发育、脊椎、肋骨发育均正常;各组胎鼠内脏观察未见异常。结论复方枸杞子口服液无明显的致畸作用,在本实验设计剂量下是安全的。

Objective To observe reproductive toxicity and teratogenicity of compound medlar oral solution in rats by intragastric administration. Method SPF Wistar rats were randomly divided into 5 groups： normal control group, high, moderate and low doses of compound medlar oral solution group, and aspirin group （positive control group）. The model groups were given a intragastric administration of compound medlar oral solution （ 10.0 mL/kg BW） ,those in the control group were treated with hi-distilled water for injection at the same volume one time per day respectively from 7 th to 16 th day after pregnancy. The pregnant rats were dissected at the 20 th day after pregnancy. Embryo and fetal developments were evaluated, external and skeletal abnormalities were observed. Result There was no significant effect on the reproductive capacity of pregnant rats. Body weight, body height and placental weight were not affected either. Breastbone, vertebra, fib ：developed well. There was no abnormality in internal organs. Conclusion According to the designed safe dosage, compound medlar oral solution had no teratogenic effect in rats.
