摘要
目的:本实验旨在采用仪器分析的手段,测定重组人红细胞生成素(rHuEPO)的唾液酸化程度。方法:利用rHuEPO中唾液酸带负电荷的特性,通过高效毛细管电泳分离了唾液酸化程度不同的rHuEPO。结果:pH4.0的磷酸盐缓冲液可有效地涂渍毛细管壁,减少电渗流,提高分辨率。在优化的测定条件下共分出了6种糖基化形式。结论:建立的HPCE法能有效地反映rHuEPO的纯度。
OBJECTIVE: To analyze the levels of Neu5Ac by instrumental methods. METHODS: High performance capillary electrophoresis was applied to separate the glycoforms derived from different levels of Neu5Ac in rHuEPO. RESULTS: The phosphate buffer of pH 4.0 could deactivate the capillary wall effectively, so that the best resolution was obtained because of the reduced electroosmotic flow. CONCLUSIONS: The HPCE method established here could be employed to effectively detect the purity, microheterogeneity and in vivo bioactivity.
