人类错配修复基因hMSH2及p27在结直肠癌及癌旁组织中的表达及意义 被引量:1

摘要 目的探讨人类错配修复基因hMSH2和p27在结直肠癌及癌旁组织中的表达及其意义。方法以免疫组化方法检测54例结直肠癌组织中hMSH2和p27蛋白的表达。结果结直肠癌组织中hMSH2蛋白的阳性表达率为70.4%,显著高于癌旁组织的38.9%(P<0.05);p27蛋白的阳性表达率为40.7%,显著低于癌旁组织的75.9%(P<0.01)。结直肠癌癌组织中hMSH2蛋白的表达与肿瘤临床病理特征无相关性;p27蛋白的表达与肿瘤分化程度、淋巴结转移和TNM分期相关。hMSH2和p27蛋白在结直肠癌组织中的表达呈负相关。结论hMSH2表达异常和抑癌基因p27表达下调可能与结直肠癌的发生有关。 Objective To investigate the expression of h MSH2 and p27 in colorectal carcinoma and paraeancerous tis- sues and their significances. Methods Immunohisto chemical method was used to determine the protein expression of hMSH2 and p27 in cancerous and paracancerous tissues of 54 eoloreetal cancer patients. Results The positivity rate of hMSH2 protein in colorectal carcinoma tissue was 70. 4% , significantly higher than that in the paraeaneerous tissue 38.9% (P〈0. 05 ), and the pos- itivitv rate of p27 protein in colorectal carcinoma tissue was 40. 7%, significantly lower than that in the paracancerous tissue 75.9% (P〈0. 05 ). Expression of hMSH2 protein in coloreetal carcinoma tissue was not relevant with the elinicopathological pa- rarneters of colorectal carcinoma, whereas expression of p27 protein was related with differentiation, invasion, lymph node metasta- sis and TNM staging of colorectal carcinoma. Negative correlation was found between hMSH2 and p27 protein expression in color- ectal carcinoma tissue. Conclusion Abnormal expression of hMSH2 and downregulation of p27 expression may have close rela- tionship with the genesis of colorectal carcinoma.
