摘要
目的:通过观察阿仑膦酸钠联合辛伐他汀对雌性去势大鼠骨质疏松的影响,来探讨两种药物联合应用治疗骨质疏松的可行性。方法:30只雌性SD大鼠随机分成6组,即去势对照组、去势阿仑膦酸钠组、去势联合用药组、假手术对照组、假手术阿仑膦酸钠组、假手术联合用药组。术后2周开始药物干预,3个月处死动物后分别测定大鼠股骨的骨密度,ELISA测定血清骨钙素(BGP)、骨保护素(OPG)、骨保护素配体(OPGL)。结果:去势对照组大鼠骨密度低于假手术对照组,去势阿仑膦酸钠组高于去势对照组(P<0.05),去势联合用药组高于去势阿仑膦酸钠组(P<0.05)。去势对照组大鼠BGP高于假手术对照组(P<0.05),去势阿仑膦酸钠组低于去势对照组(P<0.05),去势联合用药组低于去势阿仑膦酸钠组(P<0.05)。去势对照组OPG低于假手术对照组(P<0.05),去势阿仑膦酸钠组、去势联合用药组高于去势对照组(P<0.05)。去势对照组OPGL高于假手术对照组(P<0.05),去势阿仑膦酸钠组、去势联合用药组低于去势对照组(P<0.05)。结论:联合应用阿仑膦酸钠和辛伐他汀比单独应用阿仑膦酸钠对雌性去势大鼠骨质疏松有更好的防治作用,并可能通过影响OPG、OPGL发挥作用。
Objective:To observe the prevention and therapeutic effects of alendronate combined with simvastatin on the osteoporosis in rats. Methods:Thirty SD female rats were randomly divided into six groups:the ovarieetomized group (A), the OVX with alen- dronate treatment group(B), the OVX with alendronate and the simvastatin treatment group(C), the sham operation group(D), the sham operation with alendronate treatment group (E), the sham operation with alendronate and simvastatin treatment group(F). Two weeks after OVX operation, medical intervention was given into each group except of group A. After three months, all rats were sacrificed. We observed the change of rats' femoral bone density(BMD), bone gamma-carboxyglutamic-acid-containing proteins(BGP), osteoprotegerin(OPG) and osteoprotegerin ligand(OPGL). Results:The level of BMD in group A was lower than that in the group D (P 〈 0.05). The level of BMD raised in group B compared to the group A, and in group C BMD raised compared to the group B. The level of BGP in group A was higher than that in the group D (P 〈 0.05). The level of BGP decreased in group B compared to the group A, and in group C BMD decreased compared to the group B. The level of OPG in group A was lower than that in the group D (P 〈 0.05). The level of OPG increased in group B and C, compared to the group A. The level of OPGL in group A was higher than that in the group D (P 〈 0.05). The level of BGP degraded in group B and C, compared to the group A. Conclusion:Combined application of alendronate and simvastatin has better effect than simply application of alendronate on the osteoporosis in rats, and maybe take influence by OPG and OPGL.
