IFN-γ基因转染膀胱癌瘤苗诱导的特异性抗肿瘤免疫反应 被引量:2

Induction of specific antitumor immunity by interferonγ genetransferred bladder cancer vaccine

摘要 探讨转IFNγ基因膀胱癌瘤苗的可行性。采用脂质体介导法将携带鼠IFNγ基因的逆转录病毒载体转染入鼠膀胱癌细胞系BTT739中,免疫荧光法检测转基因细胞MHCI类抗原表达;动物实验观察其致瘤性和抗肿瘤免疫功能。结果显示:建立能自分泌IFNγ的膀胱癌瘤苗BTT739/IFNγ,瘤苗细胞MHCI类抗原表达量明显增加,致癌性显著降低,并可诱导机体产生特异性抗肿瘤免疫,CD+8T淋巴细胞在抗肿瘤免疫过程中起重要作用。 To study the feasibility of interferonγ genetransferred bladder cancer vaccine,retroviral vector carrying murine interferonγ (IFNγ) cDNA genes were transfected into the murine bladder cancer line BTT 739 by lipofectamine method.Expression of MHC class I antigens were detected in BTT 739 sublines through fluorometry and the tumoriginicity and the antitumor immunity were studied in the experimental models.The results demonstrated that the bladder cancer vaccine (BTT 739/IFNγ) autosecreting IFNγ has been established and the expression of MHC class I antigens has been increased in the BTT 739/IFNγ cell.It has been demonstrated that IFNγ genetransfected tumor vaccine decreased apparently the tumoriginicity and could induce the specific antitumor immunity which seems to be mediated by CD+8 effector T cells.It was concluded that the IFNγ genetransferred bladder cancer vaccine could induce the in vivo specific antitumor immunity.
