摘要
探讨转IFNγ基因膀胱癌瘤苗的可行性。采用脂质体介导法将携带鼠IFNγ基因的逆转录病毒载体转染入鼠膀胱癌细胞系BTT739中,免疫荧光法检测转基因细胞MHCI类抗原表达;动物实验观察其致瘤性和抗肿瘤免疫功能。结果显示:建立能自分泌IFNγ的膀胱癌瘤苗BTT739/IFNγ,瘤苗细胞MHCI类抗原表达量明显增加,致癌性显著降低,并可诱导机体产生特异性抗肿瘤免疫,CD+8T淋巴细胞在抗肿瘤免疫过程中起重要作用。
To study the feasibility of interferonγ genetransferred bladder cancer vaccine,retroviral vector carrying murine interferonγ (IFNγ) cDNA genes were transfected into the murine bladder cancer line BTT 739 by lipofectamine method.Expression of MHC class I antigens were detected in BTT 739 sublines through fluorometry and the tumoriginicity and the antitumor immunity were studied in the experimental models.The results demonstrated that the bladder cancer vaccine (BTT 739/IFNγ) autosecreting IFNγ has been established and the expression of MHC class I antigens has been increased in the BTT 739/IFNγ cell.It has been demonstrated that IFNγ genetransfected tumor vaccine decreased apparently the tumoriginicity and could induce the specific antitumor immunity which seems to be mediated by CD+8 effector T cells.It was concluded that the IFNγ genetransferred bladder cancer vaccine could induce the in vivo specific antitumor immunity.
