摘要
采用小鼠腹水型肉瘤(S180)细胞贴附抑制试验监测混苯作业女工浓缩尿液的体外发育毒性。分别收集接触组和对照组6例女工班末尿液100ml,用XAD-2树脂进行浓缩。另以乙醇和甘氨酸为阳性和阴性对照进行方法学验证。提示乙醇具有明显的抑制效应,呈现剂量-反应关系,甘氨酸无抑制作用;12例浓缩尿液测试结果提示接触组1例阳性,对照组全部阴性,表明现有水平的混苯接触具有潜在的生殖发育危害。参照现场调查的结果可以说明发育毒物体外筛选试验在职业人群的尿液监测中可能有着较好的应用前景。
Developmental toxicity of urine in vitro can indicate the exposure to environmentaldevelopmental toxicants and show modification effects of relevant body factor. 150-200ml urine of 6 womenoccupationally exposed to benzene/toluene/xylene (BTX) (Air concentration: benzene <4 mg/m3,toluene 30-430mg/m3 and xylene <10mg/m3) at the end of shift work,and another 6 womenunexposed to any relevant hazards as the controls were tested. The age, lifestyles,education and theirspouse of all subjects in the exposed group were similar to those of controls. 100 ml urine wasconcentrated with Amberite XAD-2 resin. Concentrated urine samples were tested with S180 cellattachment inhibition assay,which has been validated with ethanol and glycine. Among 6 urinesamples of the exposed group,one showed inhibitory effect, determined as positive,while all 6 samplesof unexposed women did not show inhibitory effect. These results showed that potentially at thepresent exposure level of BTX,the reprodustive function of exposed women and the development oftheir offsprings might be damaged. Based on existing data on the reproductive and developmentaleffects of BTX exposure,in vitro assay for screening the developmental toxicity of the concentratedurine might have a bright future in assessing the reproductive and developmental effects ofoccupational exposure to BTX.
