摘要 应用单细胞微量凝胶电泳测试技术(microgelsinglecellelectrophoresisassay,MGE)检测氯乙烯对哺乳动物肝脏细胞(肝实质性细胞和非实质性细胞)DNA损伤情况。结果发现:氯乙烯可致肝实质性细胞DNA损伤,呈剂量-效应关系,对肝非实质性细胞只有在与肝实质性细胞混合培养时才出现一定程度的DNA损伤。本文就肝实质性细胞和非实质性细胞对氯乙烯敏感性差异的原因作了讨论。结果还表明,单细胞微量凝胶电泳技术是一项灵敏的测试动物细胞DNA损伤的方法。 In order to investigate cell specific genotoxicity of vinyl chloride, rat liver parenchymal cellsand nonparenchymal cells were studied in vitro by microgel single cell electrophoresis assay (cometassay). Results showed that vinyl chloride could distinctively induce dose-dependent increase ofDNA damage in parenchymal liver cells,while in nonparenchymal liver cells only weak effects werefound after coincubated with hepatocytes. Nonparenchymal cells seemed not so sensitive togenotoxicity of vinyl chloride as parenchymal cells.
