氯乙烯致大鼠肝细胞和外周血淋巴细胞DNA损伤的活体实验研究 被引量:4

摘要 SD大鼠呼吸道染毒(1900±50)ppm氯乙烯2小时后,以肝脏灌流加酶消化方法获得肝实质性细胞和非实质性细胞,用淋巴细胞分离液分离外周血淋巴细胞,用单细胞微量凝胶电泳(MGE)技术测定上述细胞DNA损伤情况。结果发现,整体暴露氯乙烯可引起肝实质性和非实质性细胞的DNA损伤,但程度不同,且与体外试验结果相一致。氯乙烯还可引起外周血淋巴细胞DNA损伤。 Vinyl chloride is a known indirect acting carcinogen in both animal and human being,and needsto be metabolized by mixed function oxidase of the liver. In order to investigte genotoxicity of vinylchloride,rat liver cells and lymphocytes were studied in vitro by microgel single cell electrophoresisassay (comet assay)to measure the DNA effects of vinyl chloride. The results showed that vinylchloride inhaled by rats (1900±50 ppm, 2h)could induce DNA damage of liver cells andlymphocytes,and the sub-population of rat hepatocytes exists with different susceptibility to vinylchloride.
