摘要
在立体定向仪经耳去除近交系SD大鼠脑垂体基础上,用不同剂量近交新的生的Winstar大鼠制成活化脾细胞和环磷酰胺进行诱导免疫耐受性的研究。
Inbred Spraque-Dawly female rats were hypophysectomized by the sterostactic approach through eardrum. Donor spleen cells (SC) were isolated from inbred newborn Wistar rats and activated in vitro with ConA. The donor specific tolerance of the hypophysectomized rats was induced by, (1) injecting different doses of the activated spleen cells (ASC) intravenously into the recipient rats (Hpox-SD), (2) injecting cyclophosphamide (CP) intraperitoneally, (3) combined injections of ASC and CP. The result showed that the single injection of ASC or the combined injections of ASC and CP could prolong the skin allograft survival times with a mean survival time (MST) being more than 33. 5 and more than 47. 8 days respectively. The above result indicates that ASC or ASC plus CP could successfully induce the donor-specific tolerance in hypophysectomized rats.
