HPLC-ELSD法测定心安颗粒中黄芪甲苷的含量

Determination of Astragaloside Ⅳ in Xin'an Granules by HPLC-ELSD

目的：建立以高效液相色谱-蒸发光散射法测定心安颗粒中黄芪甲苷含量的方法。方法：色谱柱为Kromasil ODS C18（150mm×4．6mm，5μm），流动相为乙腈-水（35：65），漂移管温度为40℃，载气（空气）流速为2．1L·min^-1.结果：黄芪甲苷进样量在1．32～7．95μg范围内的自然对数值与峰面积积分值的自然对数值呈良好的线性关系（r=0．9989）；平均回收率为97．84％，RSD=1．01％（n=9）。结论：本法简便、准确、重现性好，可用于心安颗粒的质量控制。

OBJECTIVE： A HPLC- ELSD method was developed for determination of the content of astragaloside Ⅳ in Xin＇an granules. METHODS： The separation was performed on Kromasil ODS C18 column（ 150 mm× 4.6 mm, 5μm） with the mobile phase consisted of acetonitrile - water（35 ： 65） . The temperature of the drift tube was 40 ℃ and the flow rate of gas （air） was 2.1 L ·min^- 1. RESULTS： The linear range of astragaloside IV was 1.32- 7.95 μg （ r = 0.998 9） and the average recovery was 97.84（RSD = 1.01%, n = 9） .CONCLUSION： The method is simple, accurate and reproducible, and it can be used for the quality control of Xin＇an granules.
