输卵管妊娠中输卵管上皮Calbindin-d28k表达变化

Changes of Calbindin-d28k protein expression in tubal epitheliums of tubal pregnancy

摘要 目的:以输卵管妊娠的输卵管标本为研究对象,观察钙结合蛋白Calbindin-d28k(CaBP-d28k)在输卵管妊娠输卵管上皮组织中的表达变化,探讨CaBP-d28k与输卵管妊娠的关系。方法:30例既往月经规律、妊娠囊尚未破裂的输卵管妊娠患者,每例患者取材均包括输卵管妊娠受精卵种植部位和非种植部位的输卵管标本。经HE染色进行组织学观察确定种植部位,采用免疫组织化学法和逆转录聚合酶链反应法检测CaBP-d28k在输卵管妊娠种植部位和非种植部位输卵管上皮中的蛋白和基因表达。结果:30例输卵管妊娠非种植部位的输卵管黏膜上皮细胞均可见CaBP-d28k蛋白及CaBP-d28kmRNA强阳性表达。种植部位的输卵管黏膜上皮细胞CaBP-d28k蛋白及CaBP-d28kmRNA表达较非种植部位明显减少(P<0.05),有些种植部位CaBP-d28k蛋白表达甚至缺如。结论:输卵管妊娠种植部位的输卵管黏膜上皮细胞CaBP-d28k蛋白及CaBP-d28kmRNA表达较非种植部位明显减少,推测CaBP-d28k与输卵管妊娠的发生可能存在密切联系。 Objective: To observe the changes of Calbindin- d28k (CaBP- d28k) expression in tubal epitheliums of tubal pregnancy, explore the relationship between CaBP - d28k and tubal pregnancy. Methods : Samples of oviducts were collected from 30 cases with regular menstrual cycle and unruptured gestation sac. Both planting site of zygote and non - planting site of zygote were included, and the for- mer was detected by HE staining, the expression levels of CaBP - d28k protein and gene in planting site of zygote and non - planting site of zygote were detected by immunohistochemistry and reverse transcription polymerase chain reaction. Results: CaBP - d28k protein and gene expressed intensively in tubal epitheliums of non - planting site of zygote, which were higher than those of planting site of zygote ( P 〈 0.05 ), furthermore, some of the planting sites of zygote had no positive expression of CaBP - d28k protein. Conclusion : CaBP - d28k protein and gene expression levels in tubal epitheliums of non - planting site of zygote are higher than those of planting site of zygote, which may be related to the pathogenesis of tubal pregnancy.
