摘要
目的探讨应用甲泼尼龙对内毒素所致急性肺损伤大鼠的防治作用及可能机制。方法静注脂多糖(6 mg/kg)制成ALI模型,54只大鼠随机分为生理盐水对照组、内毒素损伤组、甲泼尼龙组(内毒素+甲泼尼龙)。采用放射免疫(ELISA)方法检测大鼠血清TNF-α、IL-8和IL-10水平,并进行肺湿/干重比(W/D)测定。结果与对照组比较,ALI组各时相点大鼠肺组织中3种炎症因子水平及W/D值均明显上升,差异有统计学意义(P<0.05),表达峰值时间:TNF-α为4 h,IL-8为2 h,而IL-10为8 h。结论甲泼尼龙可降低内毒素致大鼠ALI血清TNF-α、IL-8和IL-10水平,对大鼠ALI有防治作用。
Objective To investigate the curative effect and possible mechanism of Methylprednislone on rat model with lipopolysaccharide-induced acute lung injury.Methods The ALI model was set up by intravanously injection of lipopolysaccharide(LPS,6 mg/ kg).54 rat were randomly divided into normal saline control group,endotoxin injury group and methylprednislone group(endotoxin+methylprednislone).The changes of TNF-α,IL-8,IL-10 levels in the pulmonary tissues were detected by ELISA at different times.Besides,the lung wet/ dry weight ratio changes were observed.Results The levels of inflammatory media and W/D in ALI group were higher obviously than those in control group.The diffrences is statistical significant.The expression of TNF-α was seen to peak at 4h and IL-8 at 2h,IL-10at 8h in lung tissue after challenge.Conclusion Methylprednislone can reduce TNF-α,IL-8,IL-10 levels of the ALI serum in rat model with lipopolysaccharide-induced acute lung injury treatment,have a preventive effect against rat ALI.
