摘要
在猪口蹄疫病毒(FMDV)活性肽VP1融合蛋白基因工程菌E.coliC500构建成功的基础上,研究了C500的质粒稳定性,C500在4种不同培养基中的生长曲线,以及不同种龄、接种量、装液量、碳源、氮源对菌体生长的影响,确定了最适基本培养条件.在此基础上,进行了5L自控罐的发酵研究,通过发酵条件的改变,菌体浓度的D(600)值由4.5提高到30左右.
In this study, the fermentation conditions for E. colt C500, which was transformed with an expression constructin for VP1 of Foot-and-Mouth Disease Virus (FMDV)as a fusion protein, was studied. The studies included plasmid stability, growth curve of C500 in 4 different types of medium, and the affection of different seed-ages, amounts of seed bacteria, volume of medium in Erlenmeyer flasks, carbon sources and nitrogen sources on the growth of C500. The optimal cultivation conditions were then determined, and applied to a 5-L fermentor. The biomass of C500 was raised from 4. 5 up to 30 successfully.
