摘要
通过FLP-FRT位点特异性DNA切离和DNA定点整合技术,将酵母蔗糖酶基因(SUC2)整合到酿酒酵母染色体不同位置上,测定了SUC2基因表达情况,从而对酵母染色体位置效应进行了初步研究.发现酵母中存在染色体位置效应,但染色体位置对SUC2基因表达水平的影响并不显著.
The chromosomal position effect was tentatively investigated in Saccharomyces cerevisiae using the yeast invertase gene(SUC2). The SUC2 gene was integrated into different chromosomal loci via homologous recombination, FLP-FRT site-specific recombination and targeted integration. Its expression in the resultant transformed cell lines was then measured. The result shows that position effect exists in yeast but influences the expression of SUC2 gene quantitatively.
