辛伐他汀对肿瘤坏死因子α诱导CX3CL1表达的影响

摘要 目的通过研究辛伐他汀对肿瘤坏死因子(TNF)-α诱导血管平滑肌细胞CX3CLl表达的影响,进一步探讨辛伐他汀的抗炎机制。方法采用体外培养的SD大鼠血管平滑肌细胞;免疫荧光法检测核因子(NF)-κB的活化;RT-PCR法检测CX3CLl mRNA表达。结果 TNF-α可诱导血管平滑肌细胞NF-κB活化,并增加CX3CLlmRNA的表达;辛伐他汀可抑制TNF-α对血管平滑肌细胞NF-κB活化及CX3CLl mRNA的表达。结论辛伐他汀可能通过抑制NF-κB的活化,而下调TNF-α诱导的CX3CLl的表达。 Objective To investigate the effects of simvastatin on CX3CL1 expression induced by tumor necrosis faetor-α(TNF-α). Methods Rat Vascular smooth muscle cells (VSMCs) were cultured in vitro via nuclear factor κB (NF—κB) activation. NF—κB p65 nuclear transaction was measured by immunofluorescence. CX3CL1 mRNA expression was measured by RT-PCR. Results TNF-α activated NF—κB and induced CX3CL1 mRNA expression in rat VSMCs. Simvastatin inhibited NF-κB activation and reduced the expression of CX3CL1 in rat VSMCs. Conclusions Simvastatin downregulates the expression of CX3CL1 by inhibiting the activation of NF—κB in rat VSMCs.
