丹皮酚体外诱导间皮细胞增殖及分泌t-PA的研究

Effect of Paeonol on Proliferation and t-PA in Mesothelial Cells

目的:探讨丹皮酚影响下离体间皮细胞的增殖及分泌组织型纤溶酶原激活剂(t-PA)的情况。方法:取10只健康Wistar大鼠用于培养间皮细胞,将离体大鼠腹膜间皮细胞平均分为4组,分别加入细胞培养液RPMI-1640,RPMI-1640+TNF-α,RPMI-1640+丹皮酚,RPMI-1640+TNF-α+丹皮酚。测定各培养液中间皮细胞增殖情况及其分泌t-PA的量。结果:单独使用丹皮酚不影响间皮细胞的增殖和t-PA分泌的量(P>0.05),单独使用TNF-α可以抑制间皮细胞进入增殖状态(P<0.05),而并不影响t-PA的分泌(P>0.05),加入丹皮酚的干预后间皮细胞的增殖率明显升高,并上调t-PA的分泌(P<0.05)。结论:TNF-α可以抑制间皮细胞增殖,丹皮酚对活化的间皮细胞增殖、分泌有上调作用,丹皮酚可能在一定程度上促进损伤的腹膜早期愈合。

Objective： To study the effect of paeonol on the proliferation rate and tissue plasminogenactivator （t-PA）in mesothelial cells. Methods：Ten healthy Wistar rats were used to cultivate mesothelial cells.The cultured cells were divided into four groups,group 1 （RPMI-1640）,group 2 （RPMI-1640＋TNF-α）,group 3 （RPMI-1640＋paeonol） and group 4 （RPMI- 1640＋TNF-α＋paeonol）. The proliferation rate was detected with MTT.The level of t-PA was measured as well. Results：The proliferation rate and level of t-PA were increased in paeonol group （P 〈 0.05）.The proliferation was significantly inhibited by tumor necrosis factor （TNF-α） in mesothelial cells （P 〈 0.05）. Conclusion： TNF-ct can inhibit proliferation of mesothelial cell, but paeonol increase the proliferation and t-PA expression,It is suggested that paeonol might,in certain extent,motivate the healing of injured peritoneum.
