摘要
利用DNA条形码信息研制DNA芯片,建立快速高效的花蓟马鉴定方法。以3种花蓟马属,共9个样本的COI基因片段序列为研究材料,应用软件Primer Premier5搜索出9个样本的12种motif基因序列,应用这12种motif序列制作虚拟DNA条形码芯片。虚拟电子杂交20个样本的COI基因片段序列。结果显示,设计的虚拟DNA芯片能够鉴定本研究中的3种花蓟马,利用DNACOI基因片段,设计DNA芯片在理论上可行。
A quick method of Frankliniella identification by DNA chip that based on the DNA barcoding information was established. Nine COI gene partial sequences of three species of Frankliniella were obtained. Twelve Motif sequences of the nine samples were searched by the software Primer PremierS. Simulated DNAchip were made acorrding to the twelve Motif sequences. CO I gene partial sequences of twenty samples were hybridized with the Simulated DNA chip. It was showed that DNA barcode chip can identify the there spicies of Frankliniella. Integrating the DNA barcode and DNA chip is feasible in theory.
