灵芝多糖肽对Alzheimer样大鼠海马Aβ含量和超微结构的影响 被引量:5

摘要 目的:观察灵芝多糖肽(GLPP)对Alzheimer样大鼠海马Aβ含量及海马线粒体和神经突触的影响。方法:♂性Wistar大鼠120只,随机分为8组,每组15只:正常对照组、模型组、NS组、GLPP组(按不同剂量50mg/kg、250mg/kg和1250mg/kg分为GLPP1、GLPP2和GLPP3组)、Mel组和DMSO组;除正常对照组(正常昼夜节律)外,其余各组连续光照(光照度400Lx,24h)30d,其间GLPP组按不同剂量igGLPP,每天1次;NS组ig相同体积的生理盐水,每天1次;Mel组ipMel(1.0mg/kg),每天1次;DMSO组ip相同体积的DMSO,每天1次。光照30d后Morris水迷宫法测试各组大鼠空间记忆能力,放免法检测海马Aβ含量,透射电镜观察海马线粒体和突触结构的改变。结果:模型组大鼠寻台潜伏期明显延长,与正常对照组比较有显著性差异(P<0.05);GLPP2组、GLPP3组和Mel组寻台潜伏期明显缩短,与模型组比较有显著差异(P<0.05);GLPP1组、NS组和DMSO组寻台潜伏期明显延长,与模型组比较无显著差异(P>0.05)。模型组大鼠海马Aβ含量增多,与正常对照组比较有显著差异(P<0.05);GLPP2组、GLLP3组和Mel组大鼠海马Aβ含量减少,与模型组比较有显著差异(P<0.05);GLLP1组、NS组和DMSO组大鼠海马Aβ含量升高,与模型组比较无显著差异(P>0.05);透射电镜结果显示:正常对照组、GLPP2组、GLLP3组和Mel组大鼠海马线粒体神经轴突和神经突触基本正常;模型组、GLPP1组、NS组和DMSO组大鼠海马线粒体膜结构破坏,线粒体肿胀,线粒体嵴模糊、消失,结构紊乱,神经髓鞘内神经细丝稀少,神经突触缺失、减少,突触密度降低,突触间隙不清,突触小泡减少。结论:中剂量和高剂量GLPP可减少Alzheimer样大鼠海马Aβ含量,并可防止持续光照对大鼠海马超微结构的破坏和减轻Alzheimer样大鼠的空间记忆障碍;而低剂量的GLPP则无上述作用。
