以药效实验筛选降脂利肝颗粒的提取工艺 被引量:2

摘要 目的:通过药效实验对降脂利肝颗粒的提取工艺进行筛选。方法:通过给予高脂饮食制作高脂血症鹌鹑模型,分别给予降脂利肝颗粒水提取法、水提醇沉法、85%乙醇提取法和30%乙醇提取法4种提取工艺的浸膏,各设低、高两个剂量组共8组,另设吉非罗齐阳性对照组。检测动物血清总胆固醇(TC)、甘油三酯(TG)和低密度脂蛋白胆固醇(LDL-C)的水平。结果:水提取法和30%乙醇提取法制备的浸膏得率最高,均为32.19%。模型动物经口服给药后,动物的血脂指标均有下降,其中水提醇沉法、85%乙醇提取法和30%乙醇提取法制备的浸膏降低TC、TG和LDL-C效果较好,与吉非罗齐对照组相似。结论:经药效筛选,结合浸膏得率,确定30%乙醇提取法为4种提取工艺中的最佳工艺。 AIM: To determine the best extraction methods for the preparation of Jiangzhi pharmacodynamic research. METHODS: The quail hyperlipidemia model was constructed by Ligan Granules by feeding with high cholesterol diet flint. The model animals were treated with four different extract samples of Jiangzhi Ligan Granules, including water extraction, water extraction/ethanol sedimentation, 85% ethanol extraction and 30% ethanol extraction, each with a low and a high dosage. Serum Total cholesterol (TC), Triglyceride (TG) and Low density lipoprotein cholesterol (LDL-C) levels were used as markers for evaluation. Gemfibrozil was used as a positive control. RESULTS: The extraction rate from water and 30% ethanol was higher. Both were 32.19%. After oral ad- ministration of four different extract samples, lower serum lipid index were observed. In samples from water extraction/ethanol sedimentation,85% ethanol extraction and 30% ethanol extraction, the animal models' serum TC, TG and LDL-C levels were significantly lowered among these groups. The decrease was similar to the Gemfibrozil positive control group. CONCLUSION: After screening based on the lipid-lowering effect and the extraction rate, the extraction method used for 30% ethanol extraction is confirmed to be the best.
