针刺对单纯性肥胖大鼠脂肪组织中Bcl-2的表达及脂肪细胞凋亡的影响 被引量:6

摘要 目的通过观察针刺对单纯性肥胖大鼠脂肪组织中Bcl-2的表达及脂肪细胞凋亡的影响,探讨针刺减肥机制,为临床应用针刺减肥提供理论和实验依据。方法采用自制高脂饲料制备单纯性肥胖大鼠模型,将造模成功的20只大鼠随机分为肥胖组和针刺组,每组10只;采用普通饲料喂养的10只大鼠作为对照组。用SP免疫组织化学法测定脂肪组织Bcl-2的表达以及末端脱氧核苷酸转移酶介导dUTP缺口末端标记法检测脂肪细胞凋亡指数的变化。结果肥胖组大鼠脂肪细胞凋亡指数显著低于对照组,针刺组大鼠脂肪细胞凋亡指数较肥胖组和对照组显著升高,差别均有统计学意义(P<0.01)。肥胖组大鼠脂肪组织Bcl-2阳性水平表达率显著高于对照组,针刺组大鼠脂肪组织Bcl-2阳性水平表达率较肥胖组和对照组显著降低,差别均有统计学意义(P<0.01)。针刺单纯性肥胖大鼠时脂肪组织Bcl-2蛋白表达与脂肪细胞凋亡指数呈负相关(r=-0.88,F=8.49,P<0.01)。结论针刺单纯性肥胖大鼠时脂肪组织中的Bcl-2水平表达降低,脂肪细胞凋亡增加,可能是针刺减肥的细胞分子重要机制。 Objective To investigate the effect of acupuncture on the expression of Bcl-2 and adipocyte apoptosis in simple obesity rats, and to explore the mechanism of weight reduction function in order to provide a principle for the practice of acupuncture in clinic. Methods The model of simple obesity rats was established by high fat forage. The twenty rats with simple obesity were divided into acuouncture group and fat group. 10 in each group. The rats which were fed on normal forage were chose as control group. Expression of Bcl-2 in adipocyte tissue was determined with SP immunohistochemistry technique. The adipocyte apoptosis was determined with terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling technique. Results In obesity group, the lipoapoptosis index was lower than control group, the lipoapoptosis index in acupuncture group was higher than control group and obesity group, the differences all were statistical significance (P 〈 0.01 ). Compared with control group, the levels of expression of Bcl-2 in adipocyte tissue in obesity group was higher( P 〈 0.01 ). The levels of expression of Bcl-2 in adipocyte tissue in acupuncture group was lower than control group and obesity group ( P 〈 0.01 ). The levels of expression of Bcl-2 in adipocyte tissue of simple obesity rats was negative correlated to lipoapoptosis index ( r = - 0. 88, F = 8.49, P 〈 0.01 ). Conclusion During the acupuncture on the simple obesity rats,the levels of expression of Bcl-2 in adipocyte tissue decreased and adipocyte apoptosis increased, which may be cellular and molecular mechanisms of acupuncture for anti-obesity.
