摘要
目的探讨草氨酸盐对胰腺癌细胞panc-1糖代谢和线粒体膜电位的影响。方法体外培养人胰腺癌细胞panc-1,分别加入不同浓度的草氨酸盐培养48 h后,行酶组织化学染色及罗丹明123染色,观察细胞糖酵解关键酶乳酸脱氢酶(LDH)活性及线粒体膜电位。结果随着草氨酸盐浓度的升高,细胞LDH染色反应颗粒逐渐减少,细胞数目减少,细胞结构明显不规则;线粒体膜电位随草氨酸盐浓度增加而逐渐降低(P<0.05)。结论草氨酸盐可以抑制人胰腺癌细胞panc-1的LDH活性,阻断其糖酵解,进而影响其能量代谢杀伤肿瘤;同时可能通过降低细胞线粒体膜电位而导致细胞凋亡。
Objective To investigate the effect of oxamate on glycometabolism and mitochondrial membrance potential of pancreatic cancer cell panc-1. Methods Pancreatic cancer cell panc-1 was treated with oxamate for 48 h, the mitochondrial membrane potential was measured by flow cytometry ( Rodamin-123 ), and the activity of intracellular lactate dehydrogenase (LDH) was studied by enzymohistochemistry. Results With the increase of oxamate concentration, the mitochondrial membrane potential was decreased and the cytoplasmic granules of LDH was decreased, the number of cancer cells was decreased and the shape of the cells became irregularity, and the mitochondrial membrance potential degraded gradually( P 〈 0.05 ). Conclusion Oxamate can kill the tumor cell by restraining lactate dehydrogenase activity, obstructing the course of glucolysis and energy metabolism, and degrading the mitochondrial membrance potential.
