摘要
以11个甘蓝型油菜品种(系)的下胚轴为外植体,研究影响下胚轴细胞再生的若干因素。结果表明,用附加2,4-D的培养基对下胚轴切段进行短时间的预培养可显著提高细胞的生长和分化能力,使芽再生率达90%以上;AgNO3对细胞的生长有很大的促进作用;丝氨酸和谷氨酰胺的有益效应不明显;下胚轴的上端比下端产芽早,但两者产芽率差异不显著;不同的基因型对同一培养条件反应不同,芽分化频率差异很大。
The hypocotyl explants from 11 lines (cultivars) of Brassica napus were cultured and some factors affecting shoot regeneration were also studied. The results showed that the capacity of the growth and differentiation of hypocotyl cells was significantly increased via a preculture of 2 days period with the midium containing combination of 1.0mg/L 2,4-D and 0.2mg/L 6-BA and the shoot regeneration rate of 90.36% was obtained. AgNO3 (6mg/L)was advantageous to the shoot formation. No obvious favourable effect of Lserine (100mg/L) and Lglutamine (300mg/L) on shoot differentiation was observed, which revealed that Lserine and Lglutamine were not essential for hypocotyl culture. The upper segments of hypototyl produced shoot earlier than the lower segments but their shoot regneration frequencies were nearly same. The shoot regeneration response of 5 genotypes was assessed and the regeneration potential depended greatly on the genotypes tested.
