免疫鸡羽毛根中火鸡疱疹病毒的分离与鉴定 被引量:2

摘要 用常规剂量的商用火鸡疱疹病毒(HVT)冻干苗经腹腔免疫1日龄非免疫鸡,于第3周采集羽毛根,分别采取直接法和间接法(即经过滤获取脱细胞的游离物)接种次代鸡胚成纤维细胞(CEF),以进行病毒的分离。结果显示,在免疫后第3、4、5、6周,2种方法都分离到病毒,第7周仅直接法分离到病毒,而第8周2种方法都没有分离到。根据分离到病毒的细胞病变(CPE)特征,结合地高辛标记的Ⅰ型马立克氏病病毒(MDV-1)核酸探针检测结果,表明HVT免疫鸡羽毛根中含有可脱离细胞的、具有感染性的疫苗株HVT粒子,且其可检出时间具有阶段性。 One day old chickens without maternal antibody were inoculated intra peritoneally with turkey herpesvirus (HVT) from the commercial lyophilised vaccine, according to recommend dose of the manufacture′s instructions. Starting on week 3 post inoculation (PI) and about 1 week intervals, the virus was examined in secondary chicken embryo fibroblast (CEF) monolayer cells inoculated with the feather root preparations treated with both a direct method and an indirect method (cell free filterates). The HVT was isolated and identified on the basis of its inoculation, characteristic cytopathic effect (CPE) and hybridization of probe for serotype 1 MDV DNA,respectively. The results showed that the inoculated birds isolated at 2,3,4,5,6 weeks PI were always positive of both methods for HVT in the feather roots, whereas only the direct method was positive at 7 weeks and none at 8 weeks PI indicating that the vaccinal HVT can be detected from the feather roots of chickens at 3 to 7 weeks PI.
