8-羟基二氢小檗碱与盐酸小檗碱治疗大鼠2型糖尿病的对比研究

Comparison of Therapeutic Effects between 8-Hydroxydihydroberberine and Berberine Hydrochlorde on Type 2 Diabetes Mellitus in Rats

目的比较8-羟基二氢小檗碱(Hdber)和盐酸小檗碱(Ber)对大鼠2型糖尿病的治疗作用。方法采用小剂量链脲佐菌素(STZ)和高脂高热量饮食喂养建立Wistar大鼠2型糖尿病模型,分为正常组、模型组、Hdber低、中、高剂量组与Ber组,分别给予相应药物灌胃治疗,8周后作口服葡萄糖耐量试验(OGTT),采用葡萄糖氧化酶法检测血糖,并检测血清胰岛素(INS)、游离脂肪酸(FFA)和三酰甘油(TG)浓度。结果与正常组比较,模型组大鼠餐后血糖均明显升高,血清INS降低,FFA和TG水平升高(P<0.01);与模型组比较,Hdber中、高剂量组大鼠餐后血糖降低,FFA和TG水平降低(P<0.01),Hdber高剂量组大鼠血清INS升高(P<0.01);Hdber中、高剂量组餐后血糖与Ber组比较无明显差异,但FFA水平明显降低(P<0.01)。结论8-羟基二氢小檗碱可有效治疗大鼠2型糖尿病,其降糖降脂功效与盐酸小檗碱有一定差异。

Objective To compare the therapeutic effects between 8-hydroxydihydroberberine（Hdber）and Berberine hydrochlorde（Ber）on type 2 diabetes mellitus in rats. Methods The model of rat type 2 diabetes was established by intravenous injection of streptozotocin（STZ）and feeding with high fat and high caloric diet. Animals were randomly divided into normal group,model group, Hdber groups with different dosage and 13er group. Medications were given by intragastric administration. After 8 weeks oral glucose tolerance test（OGTT）was taken by using glucose oxidase method. Levels of serum fasting insulin（FINS ） ,free fatty acid（FFA）and triglyceride（TG）were also determined. Results Compared with normal group serum levels of postpradial glucose was increased significantly in model group. Levels of serum FINS was decreased and concentrations of FFA and TG were increased in model group（P〈0.01 ）. However,levels of blood glucose,FFA and TG were decreased in rats treated with middle and high dosage Hdber（P〈0.01） ,while levels of serum FINS in high dosage Hdber group was increased（P〈0.01 ）. There were no differences of postpradial glucose concentration between middle and high dosage Hdber groups and Ber group. Serum FFA concentrations in middle and high dosage Hdber groups were lower than that in Ber group （P〈0.01）. Conclusion It is suggested that 8-hydroxydihydroberberine is effective to be used to treat type 2 diabetes mellilus in rats. The hypoglycemic and hypolipidemie effects of 8-hydroxydihydroberberine are different with Berberine hydrochloride to a certain degree.
