密骨胶囊对去卵巢骨质疏松大鼠体内25(OH)D_3和1,25(OH)_2D_3含量及肾脏VDR mRNA表达的影响 被引量:3

摘要 目的研究密骨胶囊对去卵巢骨质疏松大鼠体内两个骨代谢指标25-羟基维生素D3[25(OH)D3]和1,25-二羟基维生素D3[1,25(OH)2D3]含量及肾脏VDR mRNA表达的影响。方法72只8月龄健康雌性SD大鼠随机分为6组,即假手术组,模型组,对照药物活性维生素D3组以及密骨胶囊小、中、大剂量组,每组12只,治疗3个月后全部处死,用双能X线骨密度仪及生物力学技术评估模型的骨密度(BMD)及骨质量,采用ELISA法检测血清和肝、肾组织中25(OH)D3和1,25(OH)2D3含量,用FQ-PCR法检测肾脏组织中VDR mRNA的表达情况。结果①模型组大鼠股骨头及粗隆部的BMD及骨生物力学指标(最大载荷和最大压应变)均明显下降(P<0.05或P<0.01);②使用密骨胶囊大、中剂量组与模型组比较,血清和肝脏、肾脏组织中25(OH)D3和1,25(OH)2D3的含量明显升高(P<0.05或P<0.01),与活性维生素D3组比较差异无统计学意义;③模型组大鼠肾脏VDR mRNA的表达低于密骨胶囊治疗组部分大鼠及假手术组(P<0.05或P<0.01)。结论密骨胶囊能促进体内VDR mRNA的表达,提高体内25(OH)D3和1,25(OH)2D3含量,从而激活骨代谢,增强骨质骨量。提示适当补充密骨胶囊可有效的防治绝经后骨质疏松。 Objective To investigate the effects of Migu capsule on the levels of two bone metabolism makers,25(OH)D3 and 1,25(OH)2D3 ,and the expression of renal vitamin D receptor(VDR) mRNA in ovariectomized rats. Methods 72 female SD rats (eight months) were randomly divided into sham group, model group, alfacalcidol treatment group,and Migu capsule groups of low,middle and high dosages. All rats were killed three months later,and then dual energy X-ray absorptiometry(DEXA) and biomechanics test were used to evaluate bone mineral density(BMD) and bone quality of the model. ELISA was used to detect the levels of 25 (OH)D3 and 1,25 (OH)2D3 in serum or tissue. While,FQ-PCR was used for quantitative analysis of expression of renal VDR mRNA. Results The BMD,index of maximal load and maximal stress of femoral head or trochanter were decreased significantly in model rats(P〈0.05 or P〈0.01),while,the levels of 25(OH)D3 and 1,25(OH)2D3 discern in serum,liver and kidney were higher in Migu capsule groups of middle and high dosages than in model group(P〈0.05 or P〈0.01). But compared with alfacalcidol treatment group, there was no significant difference. The expression of VDR mRNA were higher in Migu capsule groups of dosages than in model group (P〈0. 05 or P〈0. 01). Conclusion Migu capsule could raise the expression of renal VDR mRNA and the levels of 25 (OH)D3 and 1,25 (OH)2D3,and activate bone metabolism to reinforce quality of the bone. It indicated that appropriate amount of Migu capsule could effectively prevent and cure postmenopausal osteoporosis.
