HPV16 E7蛋白阳性和阴性乳癌组织中细胞周期调控相关基因蛋白的表达 被引量:2

Expression of cell cycle control-related gene proteins in HPV16 E7 protein-positive and-negative breast carcinoma tissue

摘要 目的:研究人乳头瘤病毒16型(HPV16)E7蛋白阳性(E7+)和阴性(E7-)乳癌组织中细胞周期调控相关基因蛋白表达的变化。方法:56例乳癌组织经PCR筛选证实HPV16 DNA阳性,其中42例HPV16 E7+,14例E7-。采用Western blotting方法检测癌组织中细胞周期调控相关基因蛋白CyclinA、CyclinD、CyclinE、P21和P27的表达。结果:42例HPV16E7+乳癌组织中,CyclinA、CyclinD、CyclinE、P21和P27蛋白阳性率分别为97.6%(41/42)、76.2%(32/42)、92.9%(39/42)、9.5%(4/42)和40.5%(17/42);14例HPV16E7-乳癌组织中,CyclinA、CyclinD、CyclinE、P21和P27蛋白阳性率分别为64.3%(9/14)、78.6%(11/14)、57.1%(8/14)、50.0%(7/14)和42.9%(6/14)。与HPV16E7-乳癌组织相比,HPV16E7+乳癌组织中CyclinA和CyclinE蛋白表达均上调(χ2分别为12.196,9.929,P均<0.01),P21表达下调(χ2=10.898,P<0.01),而CyclinD和P27的表达均无明显变化(χ2分别为0.033,0.025,P均>0.05)。结论:HPV16E7可通过统计量上调CyclinA和CyclinE表达以及下调P21表达而在乳癌发生发展中发挥重要作用。 Aim:To investigate the expressions of cell cycle control-related gene proteins in human papillomavirus 16 type(HPV16) E7 protein-positive(E7+) and-negative(E7-) breast carcinoma tissue.Methods:A total of 56 HPV16 DNA-positive breast carcinoma specimens confirmed by PCR,including 42 HPV16 E7+ and 14 HPV16 E7-,were used to examine the expressions of Cyclin A,Cyclin D,Cyclin E,P21 and P27 by Western blotting assay.Results:The positive rates of Cyclin A,Cyclin D,Cyclin E,P21,and P27 in HPV16 E7+ breast carcinoma tissue were 97.6% (41/42) ,76.2% (32/42), 92.9% (39/42), 9.5% breast carcinoma tissue were 64.3% (9/14) , respectively. Compared with those in the HPV1 were up-regulated significantly (χ2 was 12. 196 (4/42), and 40.5% (17/42), respectively, while those in HPV16 E7- 78.6% (11/14),57.1% (8/14),50.0% (7/14)and42.9% (6/14), 6 E7- breast carcinoma tissue, ,9. 929, respectively,P 〈 0.01 ) , the expressions of Cyclin A and Cyclin E the expression of P21 was down-regulated (X2 = 10. 898, P 〈0. 01 ), but the expressions of Cyclin D and P27 had not obvious changes (X2 was 0. 033,0. 025, respec- tively,P 〉 0. 05) in HPV16 E7 -breast carcinoma tissue. Conclusion: HPV16 E7 may play an important role in the carci- nogenesis and progression of breast carcinoma by up-regulating the expressions of Cyclin A and Cyclin E, and down-regula- ting the expression of P21.
