清热祛湿益气方对刀豆蛋白A诱导小鼠急性肝损伤的保护作用

摘要 目的:探讨清热祛湿益气方(QSYF)对刀豆蛋白A(ConA)诱导小鼠急性肝损伤的影响。方法:BALB/c雄性小鼠随机分为5组,空白对照组和模型对照组分别给予双蒸水20mL/kg灌服,QSYF低、中、高剂量组分别给予QSYF药液5.0、10.0、15.0g/kg灌服,连续7d。末次给药后除空白对照组外,余组采用ConA静脉注射诱导小鼠急性肝损伤模型,6h后处死动物检测血清丙氨酸氨基转移酶(ALT)、总胆红素(TBIL),肝组织超氧化物歧化酶(SOD)和丙二醛(MDA)含量,同时计算小鼠肝、脾指数和病死率。结果:5组小鼠病死率差异无统计学意义(χ2=6.579,P>0.05);5组小鼠肝、脾指数,血清ALT、TBIL水平及肝组织MDA和SOD含量差异均有统计学意义(F分别为14.560,22.842,15.522,8.201,7.755,20.952,P均<0.05)。与空白对照组比较,模型对照组肝、脾指数,血清ALT、TBIL及肝组织MDA含量均升高,SOD含量下降,差异均有统计学意义(P<0.05);与模型对照组比较,QSYF中、高剂量组脾指数、血清ALT水平降低,QSYF高剂量组肝组织SOD含量升高,差异均有统计学意义(P<0.05)。结论:QSYF对ConA诱导的急性肝损伤小鼠具有保护作用。 Aim :To investigate the protective effects of qingrequshiyiqi prescription(QSYF) on acute hepatic injury in mice induced by concanavalin A (Con A). MeIhods:BALB/c mice were divided into 5 groups, among which, 3 groups were given 5.0,10.0,15.0 g/kg QSYF by gastric perfusion respectively,and normal control group and model group were given normal saline for 7 days. After the last administration,all the groups except normal control group were established the pathologic model of acute hepatic injury induced by Con A. The contents of ALT, TBIL, SOD, MDA, the fatality rate and the weighing indexes of liver and spleen were measured. Results :The weighing indexes of liver and spleen, serum levels of ALT and TBIL and content of MDA and SOD in liver tissue among the 5 groups all had significant differences ( F = 14. 560, 22. 842,15. 522,8. 201,7. 755,20. 952, respectively, P 〈 0.05 ). Compared with normal control, the weighing index of liver and spleen,serum levels of ALT and TBIL and content of MDA in model group increased, and content of SOD decreased (P 〈 0. 05 ) ;compared with model group,the weighing index of spleen and serum ALT level in the QSYF middle and high dosage groups decreased, and content of SOD in QSYF high dosage group increased (P 〈 0. 05 ). Conclusion : QSYF has protective effects on acute hepatic injury in mice induced by Con A.
