摘要
目的:检测人乳癌组织中转移相关基因(MTA1)、基质金属蛋白酶9(MMP-9)和组织型金属蛋白酶抑制剂(TIMP-1)的表达。方法:采用免疫组织化学SP法检测56例乳癌和癌旁正常组织中MTA1、MMP-9和TIMP-1蛋白的表达。结果:乳癌组织中MTA1、MMP-9和TIMP-1蛋白的阳性表达率分别是69.6%、73.2%和51.8%,均高于正常组织中的7.1%、10.7%和5.4%,差异有统计学意义(χ2=46.24、42.38、27.34,P<0.05)。MTA1、MMP-9和TIMP-1蛋白的表达与乳癌临床分期和淋巴结转移有关(P<0.05)。乳癌组织中MTA1和MMP-9蛋白表达有关(χ2=8.151,P<0.05)。结论:联合检测MTA1、MMP-9、TIMP-1的表达可为预测乳癌浸润转移的潜能提供参考。
Aim : To detect metastasis associated gene ! ( MTA1 ) , matrix metalloproteinase 9 (MMP-9) and tissue inhibitor of matrix metalloproteinasel ( TIMP-1 ) protein in breast cancer and analysis the relationship between these proteins expression and pathobiology behavior. Methods:SP Immunohistochemical technique was used to detect the expression of MTA1, MMP-9,TIMP-1 proteins among 56 human breast cancer and 20 normal breast tissues. ReSults: The positive rates of MTA1, MMP-9 and TIMP-1 proteins were 69.6% , 73.2% and 51.8% respectively, which were 7. 1% , 10.7% and 5.4% in normal breast tissues and there were significant differences(χ2 = 46.25,42.38 and 27.34, P 〈 0.05 ). The over expression of MTAI or MMP-9 and the low expression of TIMP-1 prorein were related with degree of differentiation,clinical stage and lymph node metastasis (P 〈 0.05 ). In addition ,there was relationship between MTA1 and MMP-9 in human breast cancer(χ2 = 8. 515, P 〈 0.05 ). Conclusion : The united detection of MTA1, MMP-9 and TIMP-1 expression may predict the ability of invasion and metastasis of breast cancer.
