骨髓基质细胞移植对心肌梗塞心肌羟脯氨酸表达的影响

摘要 目的:评价骨髓基质细胞移植对组织羟脯氨酸(Hyp)含量的影响,探讨细胞移植对心肌梗塞后心室重塑机制。方法:30只日本大耳白兔,随机分为骨髓基质细胞(MSC)组、安体舒通组、对照组。体外穿刺提取骨髓,分离、培养骨髓基质细胞,MSC组、对照组心肌梗塞模型制作成功2周后沿梗塞周边区域注射骨髓基质细胞悬液(3×109/100μl)或DMEM培养基100μl,安体舒通组模型制作成功后服用安体舒通20mg。心脏超声评价心功能,测定血浆、心肌组织Hyp含量、基质金属蛋白酶(MMP-9)含量,以氯化四唑(TTC)染色评价梗塞面积。结果:与对照组、安体舒通组相比,MSC组心功能提高[LVEF:(53.22±2.13)%:(56.91±2.04)%:(62.61±2.37),P<0.05];梗塞面积下降:(29.73±2.11)%:(28.61±1.24)%:(22.82±3.12)%,P<0.05;血浆Hyp含量(μg/ml)显著下降[(40.16±2.31):(34.24±1.98):(22.79±1.69),P<0.05];心肌组织Hyp含量(μg/mg湿重)显著下降[(3.59±0.19):(2.67±0.81):(1.59±0.89),P<0.05];血基质金属蛋白酶含量(MMP-9,μg/L)显著增加[(218.64±115.23):(248.16±103.9):(719.25±312.2),P<0.05]。结论:细胞移植可抑制组织羟脯氨酸合成,促进基质金属蛋白酶表达,增加心肌胶原降解,表明干细胞移植可抑制心肌胶原合成、抑制心肌局部间质纤维化,是细胞移植心功能改善机制之一。 Objective: To assess the effect of marrow stroma cell transplantation on constitution hydroxyproline of myocardial infarction in rabbit. Methods: Isolated MSCs from 30 rabbits by bone marrow puncture were cultured in cell culture. Cell implanted on two weeks after the left anterior descending branch was ligated. Rabbits was divided into control group, antisterone group and cell implanted group. The 100μl DMEM or cell suspension was injected into the border area of myocardial infarction. The heart function was assessed by echocardiography before and after cell implanted 4 weeks. The area of infarction were estimated by TTC. The constitution hydroxyproline and MMP-9 were estimated. Results: Compared with control group and antisterone group, cardiac function ameliorated ELVEF: (53.22± 2.13)% vs. (56.91±2.04)% vs. (62.61±2.37)%, P〈0. 053; the area of infarction [ (29.73±2.11)% vs. (28.61±1.24)% vs. (22.82±3. 12)%, P〈0.05]; plasma hydroxyproline reduced [(40.16±2. 31) vs. (34.24± 1.98) vs. (22.79±1.69), P〈0.05], and constitution hydroxyproline also reduced [ (3.59±0.19) vs. (2.67± 0.81) vs. (1.59±0.89), P〈0.053; the MMP-9 increased [ (218.64±115.23) vs. (248.16±103.9) vs. (719.25±312.2), P〈0. 053 in MSC group. Conclusion: The result show that mesenchymal stem cells transplantation can obviously ameliorate cardiac function, decrease infarction area and myocardium fibrosis.
