摘要
目的:研究辛伐他汀离体情况下对人T淋巴细胞活性的影响。方法:体外培养人T淋巴细胞,用植物血凝素(PHA)作为刺激剂,加或不加辛伐他汀,分组培养。孵育24h、48h和72h后收集细胞及培养上清液,以流式细胞术测定T淋巴细胞抗原,以实时逆转录聚合酶链反应(Real-timeRT-PCR)检测T淋巴细胞肿瘤坏死因子(TNF)-α、白介素(IL)-6基因表达水平,放免法测定上清液TNF-α、IL-6浓度,免疫组化方法检测T淋巴细胞内核因子(NF-κB)的表达水平。结果:①辛伐他汀可以明显抑制人T淋巴细胞表面抗原CD25、CD69的表达(P<0.05);②辛伐他汀对人T淋巴细胞表达分泌TNF-α、IL-6有明显抑制作用(P<0.05);③辛伐他汀可以明显抑制T淋巴细胞内NF-κB的转位入核(P<0.05)。结论:辛伐他汀可以抑制人T淋巴细胞活性,可能是其抗动脉硬化,治疗心衰的机制之一。
Objective: To study the inhibitory effect of simvastatin on the activity of T lymphocy'tes. Methods: Jurkat cells were incubated with PHA with of without simvastatin in vitro, simvastatin was added on 2 h before PHA. The cells were collected after incubation 24, 48, 72 h. Then the expression of cell markers such as CD25, CD69 on cell surface was assayed with flow eytometry, and the levels of TNF-α and IL-6 mRNA were quantified with real time fluorescent quantitative polymerase chain reaction. The TNF-α and IL-6 concentrations in the supernatants of Jurkat cells were measured by radioimmunoassay, and the transportion of NF-κB into the nuclear was detected by immunocytochemistry. Results: (1)Simvastatin significantly inhibited the expression of CD25 and CD69 on T lymphocyte surfaces (P〈0. 05);(2)The expression and production of TNF-α and IL-6 were inhibited by simvastatin (P〈 0.05) ; (3)Simvastatin significantly inhibited the transportion of NF-κB into the nuclear (P〈0.05). Conclusion: Simvastatin had inhibitory effect on the activity of human peripheral blood T lymphocytes, it may be the mechanism of anti-arteriosclerosis and heart dysfunction.
