Expressions of Bnip-3,Bcl-2 in experimental diabetic cardiomyopathy

摘要 目的探讨Bnip-3、Bcl-2在实验性糖尿病心肌病中的表达。方法16只雄性成年Wistar大鼠随机分为糖尿病组(10只)与对照组(6只),糖尿病组一次性腹腔注射链脲佐菌素(STZ)溶液诱导建立糖尿病模型,对照组腹腔注射枸橼酸缓冲液。12周后处死大鼠,取血检测血脂、心肌肌钙蛋白-I,切取心室肌组织HE染色观察,并随后行免疫组化染色,观察Bnip-3、Bcl-2的表达情况。结果血生化指标及心肌形态学改变均符合糖尿病心肌病表现。免疫组化染色显示Bnip-3主要阳性表达部位为细胞核和细胞质,糖尿病组Bnip-3阳性表达率明显高于对照组〔(57.70±12.77)%vs(36.68±5.86)%,P<0.01〕;Bcl-2主要阳性表达部位为细胞膜和/或细胞质,糖尿病组Bcl-2阳性表达率明显低于对照组〔(22.34±12.70)%vs(74.03±10.45)%,P<0.01〕。结论Bnip-3在糖尿病心肌细胞中表达增强,具有促进心肌细胞凋亡的作用;Bcl-2呈现弱表达,其抑制心肌细胞凋亡作用减弱,二者均加速了心肌细胞的凋亡。 Objective To explore expressions of Bnip-3, Bcl-2 in experimental diabetic cardiomyopathy (DC) model. Methods Sixteen adult male Wistar rats were randomly divided into diabetes group (n = 10) and control group (n =6). Wistar rats of diabetes group were induced to diabetes by peritoneal injection with streptozotecin(STZ) solution,and Wistar rats of control group received equal volume citric acid buffer solution peritoneal injection. After 12 weeks, all rats were killed. Lipid profile, bcod glucose and cardiac tropnin-I (cTnI were detected; HE staining was performed to observe morphologic changes of ventricular muscle tissue; immunohistochemistry staining was performed afterwards to observe the expressions of Bnip-3 and Bcl-2. Results The changes of blood chemistry indexes as well as myocardial morphologic changes all conformed to DC. Bnip-3 positive expression was located in the cytoplasm and nucleelus, and its positive expression ratio in diabetes group was significantly higher than that in control group [ ( 57.70 ± 12. 77 ) % vs ( 36. 68 ±5. 86 ) %, P 〈 0. 01 ]. The positive expression of Bcl 2 was located in the cell membrane and/or cytoplasm, and its positive expression ratio in diabetes group was significantly lower than that in control group[ (22.34±12.70)% vs (74.03±10.45)%,P〈0.01]. Conclusions The significantly increased expression of Bnip-3 and decreased expression of Bcl-2 show that Bnip-3 and Bcl-2 might cooperate in accelerating cell apoptosis.
