摘要
目的探讨激素性股骨头缺血坏死的发病机理及活血化瘀法防治的作用机制。方法通过大剂量激素诱导体外培养的骨髓间充质干细胞(BMSCs)成脂分化,在诱导成脂的同时给予桃红四物汤含药血清干预。检测干预6 d后细胞内成骨标志物I型胶原mRNA及碱性磷酸酶的表达。结果活血化瘀中药可对抗激素诱导下的BMSCs内碱性磷酸酶含量减少及I型胶原mRNA表达的降低。结论活血化瘀类中药防治激素性股骨头缺血坏死的机理不仅是改善股骨头的微循环,同时还与其抑制激素诱导下的BMSCs成骨分化减少有关。
Objective To investigate the pathogenesis of hormonal avascular necrosis of femoral head and the therapeutic mechanism of the method of activating blood and removing stasis. Methods The differentiation of bone marrow stromal cells (BMSCs) into adipocytes in vitro were induced by large dose of steroid and treated simultaneously Taohong Siwu decoction drug-containing serum for 6 days, then the expressions of osteogenic markers, including type I collagen mRNA and alkaline phosphatase, were detected. Results Huoxue Huayu decoction could antagonize the decrease of expressions of type I collagen mRNA and alkaline phosphatase in steroid-induced BMSCs. Conclusion Huoxue Huayu decoction can improve minicirculation of femoral head and inhibit the decrease of osteogenic differentiation of BMSCs induced by steroid.
