同源盒基因B7在乳腺癌组织中的表达及其临床意义

Expression of homeobox B7 in breast cancers and its clinical significance

摘要 目的探讨同源盒基因B7(homeobox B7,HOXB7)在乳腺癌组织中的表达及其意义。方法应用实时荧光定量逆转录聚合酶链反应(Real-time RT-PCR)方法检测20例乳腺癌及其配对正常乳腺组织中HOXB7 mRNA的表达;应用免疫组织化学SP法检测57例乳腺癌及其配对正常乳腺组织中HOXB7蛋白的表达水平。结果乳腺癌组织和正常乳腺组织中HOXB7基因扩增的△Ct值分别为(3.58±1.44)和(6.47±1.27),正常乳腺组织中HOXB7 mRNA表达明显低于乳腺癌组织(P<0.001);HOXB7蛋白在乳腺癌组织的表达水平明显高于正常乳腺组织(P<0.001),淋巴结转移阳性的乳腺癌病例中HOXB7的表达水平高于淋巴结转移阴性组,两者差异具有统计学意义(P=0.043)。结论HOXB7高表达可能促进了乳腺癌的发病过程,并与乳腺癌淋巴结转移相关。 Objective To investigate the correlation of the expression of homeobox B7 (HOXB7)and the occcurrenee of human breast cancer. Methods The expression of HOXB7 mRNA in 20 human fresh breast cancer eases and the corresponding adjacent normal mammary tissues was detected by real-time reverse transcriptase-polymerase chain reaction (RT-PCR),and the expression of HOXB7 protein in 57 human breast cancer cases and the corresponding adjacent normal mammary tissues was determined by immunohistochemistry. Results The mean ACt of amplification for HOXB7 gene was 3.58 (sd=1.44)and 6.47 (sd=1.27)in cancer tissues and benign mammary tissues,respectively. The expression of HOXB7 mRNA was higher in breast cancer than that in normal mammary tissues (P〈0.001). Compared with primary breast cancers, a significant reduction in expression of HOXB7 protein was noted in benign mammary lesions (P〈0.001). Moreover,the expression of HOXB7 protein was markedly increased in breast cancers with lymph node metastasis compared with without metastasis (P=0.043). Conclusion Increased expression of HOXB7 may play a role in the pathogenesis and metastasis of breast cancer.
