摘要 目的观察穴位埋药线对脑缺血再灌注大鼠脑皮质区基质金属蛋白酶-9(MMP-9)的表达和含量的影响,探讨穴位埋药线对局灶性脑缺血再灌注损伤的保护机制。方法将42只SD大鼠随机分为正常组、模型组(1d组、3d组、5d组)、穴位埋药线组(1d组、3d组、5d组)。采用改良线栓法制备局灶性脑缺血(MCAO)再灌模型,在"大椎"、"百会"穴埋入胶原川芎嗪缓释剂药线。蛋白免疫印迹法检测缺血侧脑皮质区组织MMP-9蛋白的表达与含量。结果正常组可见少量MMP-9表达,模型组MMP-9表达明显增高,与正常组有显著性差异(P(0.01);穴位埋药线组MMP-9表达与模型组各相同时相组比较,有显著差异(P(0.01),并随时间的推移趋于正常。结论穴位埋药线疗法可明显下调脑缺血再灌注大鼠缺血侧皮质组织MMP-9的表达,减少脑微血管基底膜的降解,保护微血管的完整性,减轻脑缺血再灌注损伤,起到脑保护作用。 Objective To observe the effects of embedding medicine and catgut in acupoints on the expression and contents of Matrix Metalloproteinase -9 (MMP- 9) in cortex tissues of rats with focal cerebral ischemia reperfusion injury, and discuss the protective mechanisms of embedding medicine and catgut in Acupoints against the focal cerebral ischemia reperfusion injury. Methods 42 SD rats were randomly divided into normal group, model groups (subgroups: 1d, 3d, 5d), Acupoints embedding groups (subgroups: 1d, 3d, 5d). The modified method of suture embolism was used to produce middle cerebral artery occlusion (MCAO) and reperfusion models. The collagen and Tetramethylpyrazine contained sustained release preparation was embedded into" Baihui" ( GV 20 ) and" Dazhui" ( GV 14 ) Acupoints for the Acupoints Embedding groups. Western blotting was used to determine the expression and content of MMP -9 in ischemic cortex tissues. Results The contents of MMP - 9 in cortex tissues of model groups were significantly higher than that of normal group, which had few expression of MMP - 9 (P 〈 0.01 ). Comparisons between Acupoints Embedding groups and model groups at the same time phases showed significant difference (P 〈 0.01 ), and showed an inclination to normalize the MMP -9 content with the time. Conclusion The method of embedding medicine and catgut in acupoints could obviously down - regulate the expression of MMP - 9 in cortex tissues of rats with focal cerebral ischemia reperfusion injury for cerebral protection via reducing cerebral microvascular basement membrane degradation to ensure the integrity of microvessels and at-tenuate cerebral ischemia reperfusion injury.
